I created a video demonstrating scale calibration using a calibration slide.

The video starts by showing a scale calibration which is a long way from true. It then quickly demonstrates the idea behind the scale calibration. Two points on the same image are selected, where we know the real world distance between them; in this case it’s ½ mm or 500 microns. Before calibration it shows as an grossly incorrect value, for demonstation purposes.

Note: the distance between the two selected points is shown half way along the line. Do not confuse with the cursor coordinates shown at the cursor location.

The video shows.

  • The scale bar shows 20 microns
  • A measured distance is incorrect
  • The calibration tool is selected
  • An image is captured
  • Two points are selected in the image, giving us a pixel distance between them
  • The calibrated distance is entered
  • Once calibrated the scale bar shows 100 microns
  • A measured distance is now correct

Note that for the sake of brevity in the video, no attempt has been made to click with great precision. The video simply demonstrates the software.
